Sometimes I close the ultraedit by mistake when I was just intended to lose one file since the two close button are close. Is is possible to get a warning when close multiple files?

No, there is no warning. You can customize the toolbar and menus. So if you often press the wrong close function by mistake, move the Close All Files menu item or toolbar symbol to a different location with some space between the Close File command or use hot keys.
